Research context

Melanotan II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone (alpha-MSH) that is constrained by an internal lactam bridge. It is a non-selective melanocortin-receptor agonist within the melanocortin peptide class. In vitro and animal models describe it as a ligand that engages multiple melanocortin receptor subtypes, including MC1R, MC3R, and MC4R.

Research in isolated cell and rodent models has examined melanotan II in relation to melanocortin-receptor binding and downstream cAMP/PKA signaling, as well as melanogenesis via the tyrosinase pathway. Analytical characterization has used reversed-phase HPLC and mass spectrometry to confirm the cyclic structure, lactam-bridge closure, and purity of preparations.
