Research context

GHK-Cu is a copper(II) coordination complex of the naturally occurring tripeptide glycyl-L-histidyl-L-lysine (Gly-His-Lys). The peptide chelates a single divalent copper ion, principally through the histidine imidazole nitrogen and backbone amide/amine nitrogen donors, forming a defined 1:1 metal-peptide complex. In vitro and in animal models it has been characterized within the copper-binding peptide chemical class and studied as a copper-delivery ligand interacting with extracellular-matrix components and matrix metalloproteinases.

Research in isolated cell cultures and rodent models has examined GHK-Cu in relation to copper ion transport, fibroblast gene-expression profiles, and signaling associated with collagen and glycosaminoglycan turnover. Analytical characterization has employed reversed-phase HPLC, UV-Vis spectroscopy, and mass spectrometry to confirm complex stoichiometry, purity, and the copper coordination geometry, and to measure binding constants in buffered systems.
