Research context

NAD+ (beta-nicotinamide adenine dinucleotide, oxidized form) is a dinucleotide coenzyme composed of an adenine-containing nucleotide and a nicotinamide-containing nucleotide joined through a pyrophosphate bridge; it is a coenzyme rather than a peptide. In vitro and in animal models it has been characterized as a redox cofactor and co-substrate that participates in enzymatic electron-transfer reactions and serves as a substrate for NAD+-consuming enzymes such as sirtuins and poly(ADP-ribose) polymerases (PARPs).

Studies in isolated cells and rodent models have examined NAD+ in the context of central metabolic pathways, including glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation, as well as signaling by NAD+-dependent enzymes. Analytical characterization commonly employs HPLC, LC-MS, and UV-spectrophotometric methods to quantify NAD+ and to distinguish it from its reduced (NADH) and phosphorylated (NADP+) counterparts.
