Research context

MOTS-c (mitochondrial open reading frame of the 12S rRNA type-c) is a 16-residue mitochondrial-derived peptide encoded within the mitochondrial 12S rRNA region. In vitro and in animal (rodent) models it has been described as interacting with the AMP-activated protein kinase (AMPK) signaling axis and, under metabolic stress, translocating to the nucleus where it has been reported to associate with stress-response transcription-factor pathways such as NRF2/antioxidant-response elements.

Research in isolated cell cultures and rodent models has examined MOTS-c in relation to cellular energy-metabolism pathways, including AMPK activation, folate-methionine cycle intermediates, and expression of glucose-handling genes. Analytical characterization of synthetic MOTS-c preparations has been reported using reversed-phase HPLC for purity assessment and mass spectrometry (ESI-MS and MALDI-TOF) for confirmation of the peptide molecular weight and sequence.
