Semaglutide

Semaglutide is a 31-amino-acid analog of human glucagon-like peptide-1, a gut-derived incretin. Three engineered modifications distinguish it from the native sequence. An alpha-aminoisobutyric acid substitution at position 8 blocks the dipeptidyl peptidase-4 cleavage site that degrades native GLP-1 within minutes. A C18 fatty diacid chain is conjugated at lysine 26 through a gamma-glutamate and two 8-amino-3,6-dioxaoctanoic acid spacers. Arginine replaces lysine at position 34 to direct that acylation to a single site.

The fatty diacid moiety drives reversible binding to serum albumin, which is the principal determinant of the extended circulating half-life reported for the molecule in the literature. Its target, the GLP-1 receptor, is a class B G-protein-coupled receptor expressed on pancreatic beta cells and in hypothalamic and hindbrain nuclei including the arcuate nucleus and area postrema.

In isolated islet and INS-1 beta cell preparations, GLP-1 receptor agonism couples through Gs to adenylate cyclase, raising intracellular cAMP and activating protein kinase A and the guanine nucleotide exchange factor Epac2. The resulting potentiation of insulin secretion is glucose-dependent in these models, which is a defining feature of the incretin class in vitro.

Animal-model work has examined receptor-binding kinetics, gastric emptying rate, and food intake in relation to signalling at hypothalamic and brainstem GLP-1 receptor populations. Radioligand displacement and cAMP accumulation assays in receptor-expressing cell lines are the standard in-vitro systems for characterising analogs of this class, and albumin-binding affinity is measured separately by surface plasmon resonance, since it rather than receptor affinity governs the extended persistence of the acylated sequence.

Each lot is released against reversed-phase HPLC with ultraviolet detection for purity and electrospray ionisation mass spectrometry for identity, and the results are reported on the lot-specific Certificate of Analysis supplied with the order.

For this sequence the analysis that carries the most weight is verification of the acylation site. Misacylation at an alternative lysine produces a molecule of identical mass, so mass spectrometry alone cannot distinguish it from correctly assembled material; only chromatographic separation or peptide mapping after enzymatic digestion resolves it. Related-substance profiling covers the deletion sequences and truncated fragments left by incomplete coupling during solid-phase synthesis. Because the acylated sequence is long and strongly hydrophobic, those impurities elute close to the main peak, and separating them is the practical ceiling on achievable purity for this compound.
