Retatrutide

Retatrutide is a 39-residue synthetic peptide that engages three distinct class B G-protein-coupled receptors: the GIP receptor, the GLP-1 receptor, and the glucagon receptor. Like tirzepatide it is built on a GIP-derived backbone, and it carries alpha-aminoisobutyric acid substitutions that block dipeptidyl peptidase-4 cleavage, together with a C20 fatty diacid conjugated through a spacer to drive albumin binding.

Adding glucagon receptor engagement to an incretin backbone is the design problem this molecule represents. The three receptors share structural family but differ in their orthosteric pockets, so the sequence must satisfy three binding requirements simultaneously, and the resulting potency ratio across the three is the defining characteristic reported for any given analog in this class.

Characterisation requires three parallel cAMP accumulation assays, one per receptor, in separate cell lines each expressing a single target. Reporting relative potency across those three systems is the standard way this compound class is described, and the ratio rather than any single potency figure is what distinguishes one triple agonist from another.

Glucagon receptor agonism is the component that most differentiates the reported in-vitro profile from dual agonists, since hepatocyte-model work examines cAMP signalling and downstream gluconeogenic and lipolytic pathways that incretin receptor engagement alone does not reach. Animal-model studies have examined receptor occupancy and energy expenditure endpoints in relation to signalling at all three receptor populations.

Purity by reversed-phase HPLC and identity by mass spectrometry are reported per lot on the Certificate of Analysis. As with any long acylated sequence carrying sterically hindered residues, incomplete coupling is the principal synthesis-related risk, and the resulting deletion sequences elute close to the main peak on a 39-mer.

Verification of the acylation site matters here for the same reason it does across this class: a fatty chain attached at the wrong lysine yields an identical molecular mass and is invisible to mass spectrometry alone. Because three receptor affinities depend on the intact sequence, chromatographic resolution of closely eluting related substances is the practical determinant of usable purity.
