A four-component preparation adding KPV to the GHK-Cu, BPC-157 and TB-500 combination. The added tripeptide is the one most likely to be missed by a routine chromatographic method.
KLOW Blend is a physical mixture of four separately defined compounds: KPV, the tripeptide Lys-Pro-Val corresponding to the C-terminal three residues of alpha-melanocyte-stimulating hormone; GHK-Cu, the copper(II) complex of glycyl-L-histidyl-L-lysine; BPC-157, a fifteen-residue synthetic peptide; and TB-500, a peptide corresponding to the actin-binding region of thymosin beta-4.
As with any blend it has no CAS number, no molecular formula and no molecular weight of its own. It differs from the three-component preparation by the addition of KPV, which is a considerably smaller molecule than the other three and acts on an unrelated pathway, so it is a genuine fourth entity rather than a variation on the others.
KPV is notable within the mixture for what it lacks. It carries no copper, no cysteine, no aromatic residue and no terminal modification, which makes it the simplest component chemically and, as set out below, the most awkward one to detect.
No primary literature addresses this four-way combination. Each component has its own separate evidence base, and the composite is an inference rather than a finding.
KPV is studied in cultured intestinal epithelial and immune cell lines, with nuclear factor kappa B pathway activation and cytokine transcript levels as the usual endpoints. GHK-Cu appears in fibroblast matrix and copper-transport work, BPC-157 in angiogenic signalling models, and TB-500 in actin sequestration and cell migration assays. Four unrelated molecular targets means four unrelated literatures, and predicting an interaction between them from published single-compound work is not supportable.
The chromatography here has a specific pitfall. KPV has a molecular weight near 342 daltons and is highly polar, so on a reversed-phase column it elutes very early, close to the void volume, where injection artefacts and unretained salts also appear. A gradient developed to resolve the three larger components will often push KPV into that region and lose it, and the resulting chromatogram looks clean while quietly omitting one of the four components.
A method that genuinely characterises this blend therefore starts at low organic content, or uses a stationary phase suited to polar analytes, so that the tripeptide is retained and integrated rather than discarded with the solvent front. Mass spectrometric confirmation of each of the four peaks, plus copper content and stoichiometry for the GHK-Cu component, completes the picture. As with any mixture, a single aggregate purity figure conveys very little.
