Research context

A hexapeptide ghrelin receptor agonist, less selective than ipamorelin by design and by reported profile. Supplied lyophilised, with C-terminal amidation verified per lot.

Structure and molecular target

GHRP-2, also known by the international nonproprietary name pralmorelin, is a hexapeptide with the sequence D-Ala-D-2-Nal-Ala-Trp-D-Phe-Lys and a C-terminal primary amide. Three of its six residues are D-configured or unnatural, which is what allows so short a sequence to resist peptidase cleavage.

It agonises the growth hormone secretagogue receptor 1a, the same target as ghrelin and ipamorelin. Unlike the GHRH receptor engaged by sermorelin and the CJC-1295 series, this receptor signals principally through Gq, so the appropriate in-vitro readouts are calcium mobilisation and inositol phosphate accumulation rather than cAMP.

Findings in the published research literature

The comparative literature between this compound and ipamorelin is the most informative body of work for placing it. Both engage the same receptor, but GHRP-2 is reported in animal models to elevate adrenocorticotropic hormone, cortisol and prolactin alongside its primary effect, whereas ipamorelin was specifically characterised as producing markedly less of that cross-activity at comparable receptor occupancy.

That difference is the reason both compounds remain in use as research articles rather than one superseding the other: a study examining secretagogue signalling in isolation and a study examining the broader endocrine response require different tools. Pituitary cell preparations are the standard ex-vivo system for both.

Analytical verification

Reversed-phase HPLC establishes purity and mass spectrometry confirms identity, reported per lot on the Certificate of Analysis. As with any D-configured sequence, epimerisation is the impurity that mass spectrometry cannot see: a D-to-L inversion at any of the three affected positions yields a diastereomer of identical mass, separable only chromatographically.

C-terminal amidation requires separate confirmation, since the free-acid by-product differs by one dalton and is substantially less active at the receptor. The single tryptophan is an oxidation liability, adding 16 daltons and typically resolving as a distinct peak, and its level on incoming documentation reflects storage and handling history as much as synthesis quality.
